The conferences cover a wide range of topics, far beyond the role of transporters in Drug-Drug Interactions, which other transporter meetings mostly focus on. The Meet the Experts Transporter conferences are relatively small meetings with 70-100 participants, and offer plenty of opportunities for networking with colleagues in the field or personal interaction with the speakers. Being held in Budapest, the 2018 spring meeting also provides a unique opportunity for you to meet SOLVO’s scientists, project managers and study directors.

"Update in In Vitro Liver System: Cells to Organoid. Application in Drug Metabolism and Pharmacokinetics (DMPK) and Toxicology"

 
Abstract : 
Biopredic this year will host its session in the Paris area, at Paul Brousse hospital. Our host there will be the Cellspace society which mission is to develop the bioprinting technologies in the field of regenerative medecine. The lineup of speakers and providers will include scientists involved in the 2D and 3D printing or fabrication of hepatic cell systems. The cell bricks are the hepatocytes, the primary and the HepaRG, and all cells ftom the non parenchymal fraction. A large part of the session will be devoted to the applications of those systems and will cover pbpk modeling, the fm estimation, the low clearance compounds, the genotox and cholestasis prediction.

Inauguration of the IC-3Rs

Biopredic International was very happy to be the sponsor of the inaugural symposium of the IC-3Rs (INNOVATION CENTRE 3R ALTERNATIVES) on September 2017.

Over the last 25 years, the team of In Vitro Toxicology and Dermato-cosmetology (IVTD), under the direction of Prof. Vera Rogiers at the Vrije Universiteit Brussel (VUB), has spearheaded the development of alternative methods that reflect the 3Rs principle for toxicology and safety applications. With the formation of the Innovation Centre-3Rs (IC-3Rs) they aim to increase the visibility of alternative methods by enhancing communication, building the local network and supporting the development of in vitro methodologies, throughout the Belgian region and Europe. 

Inaugural symposium: 25th September 2017 

To kick-off IC-3Rs, an inaugural symposium was held, with a focus on the alternative methods developed at the VUB, Jette. Cross disciplinary talks, from key VUB experts, highlighted how cutting edge research can work hand-in-hand with alternative strategies – delivering high quality and relevant science in line with a humane approach.

 

 

 

North American Meeting ISSX Meeting - September 2017

 

The 21th North American ISSX Meeting will take place in Rodhes Island on the 26th to the 29th of June 2017. Our Team will be there to represent Biopredic International.

During this meeting David Steen will present a lecture about Direct and Quantitative Evaluation of CYP450 Contribution (fm) to Drug Clearance Using the in vitro Mod-el Silensomes™.

The purpose of this presentation is to understand the role of the Silensomes™ to evaluate in vitro the fraction of the drug metabolized (fm) by each CYP in order to predict DDI. 

Regulators, including the U.S. Food and Drug Administration (FDA) have taken a prospective approach to the problem of drug interactions by assessing the interaction potential at an early stage of drug de-velopment. As a result, prediction of metabolically-based DDIs from in vitro and in vivo data has be-come a need in drug development. However, the accuracy of predicted results depends on the quality of the input dataset. In addition it is crucial that the complexities of the DDI process, as much as possible, are modeled in a well-defined algorithm based on fully reliable data.

In vitro identification and measurement of the contribution (fm) of the major cytochrome P450 enzymes (CYP450s) involved in the metabolism of a new drug candidate, also called “CYP450 phenotyping”, helps to predict the impact of other co-administered drugs (perpetrators) on the pharmacokinetics of the new chemical entity (NCE = victim). Today, a battery of in vitro tests (recommended by the regulatory agencies) is required for CYP450 phenotyping, each of which has a number of limitations.

A new in vitro model, called Silensomes™, has been developed to address the disadvantages of the current methodologies. Silensomes™ are human pooled HLM chemically and irreversibly inactivated for one specific CYP450 activity (1A2, 2A6, 2B6, 2C8, 2C9, 2C19, 2D6, 3A4) using a mechanism based inhibitor (MBI). Examples of fm evaluation for NCE metabolized by one or several CYPs will be shown. Correlation between in vitro and in vivo fm evaluation will be demonstrated

Learning objectives of this session include evaluation of fm for the DDI prediction using a new in vitro tool, SilensomesTM.

Inauguration of a new joint laboratory with Cellenion

This Wednesday, September 13, Biopredic International and Cellenion are pleased to organize the inauguration of their joint laboratory on the site of the Paul Brousse Hospital in Villejuif.

Biopredic produces cells for industrial research applications, mainly for pharmacokinetics, toxicology and sometimes for pharmacology. Cellenion, on the other hand, offers solutions and automated controlled cell distribution for applications in the fields of bio-printing and single-cell isolation.

These two companies collaborate with several research units within the iLite project, whose goal is to develop a bioartificial liver, liver on chip and, in the longer term, a transplantable bio-constructed liver through tissue engineering techniques.

The complementarity of these two companies is evident, and their implantation within the Paul Brousse hospital will both accelerate research within the Ilite consortium and establish new collaborations with researchers and clinicians in the university hospitals the region.

The iLite project: Innovations for Liver Tissue Engineering

The goals of this project are to build an external bioartificial liver, a liver-on-chip as well as a bioengineered liver. In the present project, bioconstruction of the liver will be performed by the assembly of building blocks –liver organoids, vascular networks and biliary networks- built separately and then integrated into a scaffold. Vascular and biliary networks will be produced by a mix of technologies comprising organotypic culture, micropatterning and stereolithography.

A French governmental financement

This project is financed by 3 French entities : "Ministère des Affaires sociales et de la Santé", "Ministère de l'éducation nationale, de l'enseignement supérieur et de la recherche" and "Commissariat général à l'investissement". 10 winners, including iLite project, has been selected. On the 78,4 millions euros invested, the iLite project received 8,5 millions euros. The iLite project is driven by the DHU HepatInnov and gathers several teams: "hôpital Saint-Louis" (AP-HP), "Université de Paris Saclay", the CEA, INSERM, INRIA and for companies including Biopredic International. A 5 years project

By the end of the 5-year project, the goal is to provide a prototype of an external bioartificial liver to be used for phase I and II clinical studies as well as a prototype liver-on-chip device ready for use by pharmaceutical companies producing new drugs. All steps in the project will take into consideration GMP procedures.

Title: A New Pathway and Related Assay Systems and Biomarkers for the Study and Prediction Of Cholestasis Side Effects Xenobiotics

Speaker: Joseph Antoun (Scientific Adviser - Research Instructor of Medicine at Vanderbilt University)

Abstract: Our lecture will rely on two papers published that year and that explain that the bile canaliculi contraction and the RhoK and the MLCk enzymes regulating the level of phosphorylated light chain myosin are the targets of cholestatic compounds.

BIOPREDIC - HOST:
  • 22nd-23rd June 2017 - We will host a special training session about hepatotoxicity: "Update on the hepatotoxicity AOP landscape and on the ADMET field [SEASON 2]"
Abstract : 
The relevant systems and biomarkers for adressing hepatotoxicity and DDI issues will be the focus for this training. Prediction of hepatotoxicity remains a challenge nowadays. The session will cover those new systems that are commercially available together with their relevance for predicting issues related to Cholestasis, genotoxicity. Participants will be trained on 3 models with various cell systems and Silensomes (fm (fractions metabolized) evaluation).
